Human Health – Cardiovascular Disease (CVD)

Cardiac troponin I (cTnI) is a highly specific biomarker and the gold standard for CVD diagnosis. Different anti-cTnI recombinant antibody formats –single-chain fragment variable (scFv), single-chain antibody fragment (scAb) and fragment antigen binding (Fab)– have been produced to this target. Panels of monoclonal antibodies have also been generated against cTnI 1. In order to generate anti-cTnI scFvs with improved specificity, affinity and sensitivity, several factors need to be considered:  autoantibodies to cardiac troponins, cross-reaction with skeletal TnI (sTnI), TnC and TnT, auto-phosphorylation of cTnI, proteolytic degradation of cTnI Immunogenicity, parker hydrophilicity and ideally no cysteine should be present in peptides for immunisation, as it may lead to the formation of disulfide bonds and improper folding which may cause loss of epitope conformation. After considering these factors, a total of five epitopes, cTnI23 (Ser23-Ala29), cTnI26 (Arg26-Lys35), cTnI34 (Ala34-Lys37), cTnI169 (Arg169-Glu178) and cTnI190 (Trp190-Ala196) were selected, and the generation of scFvs against these five epitopes is in progress.

A scFv recombinant antibody was also produced against the cardiac biomarker heart-type fatty acid binding protein (hFABP) 2. Antibodies have also been produced for Myeloperoxidase (MPO) and C-reactive protein (CRP).


Human Health – Prostate Cancer (PCa)

Prostate Specific Antigen (PSA) is a valuable marker for PCa diagnosis and follow up.  PSA exists bound to other proteins or in a free form. The free to total (free plus complexed PSA) PSA ratio (%free PSA) is associated with increased risk of PCa, and can be used to reduce unnecessary biopsies in patients within the diagnostic ‘grey zone’ (4–10 ng/mL total PSA). AbYBiotech has developed highly specific scFv and scAb antibodies to the free isoform, which have proved useful in immunosensor development 3,4.


Food Safety – Mycotoxins

Mycotoxins are low molecular weight molecules that contaminate food and represent a major health hazard worldwide. We have developed new specific antibodies with greater sensitivity to one of the most serious of these contaminants: Aflatoxin B1 5.


Marine Biotoxins – Microcystin

Phycotoxins are marine biotoxins produced by microalgae that enter the human food chain by being ingested by shellfish and thus pose a major threat to human health. AbYBiotech has produced scFv recombinant antibodies against microcystin that offer high specificity, sensitivity and stability 6.
